Ok, so let me back up for a moment.
I installed this plugin and when I attempted to activate it, I received “Error 500 Internal server error“.
Should I begin to deactivate existing plugins and attempt to re-activat Role Scoper or not?
Please advise at your earliest opportunity.

UPDATE #1 Plugin is now activated successfully. I de-activated the MiniMeta Widget.
UPDATE #2 Plugin is activated and I can partially navigate through some of the plugins options when all of a sudden, the Error 500 message appears again.
Any ideas would be greatly appreciated.

I'd be really shooting in the dark on this unless I at least knew the list of plugins you're using. A copy of your php error log and .htaccess file could be very helpful too.
If you e-mail me a login I can take a look at your configuration and maybe insert some diagnostic output into the plugin.

wp-postratings 1.31 also causes this error for me. I am hosted on 1and1 with sql5… not sure if the latter information is relevant, but that plugin definitely kills the entire admin section (the front end continues to work as far as I can tell)

Flutter .14 (formerly freshpost) also causes this error 500. If you need a login to look around please let me know.

Do you mean WP-postratings and Flutter work fine unless they're activated with Role Scoper, then you get the 500 error?

Are you still seeing a conflict with Flutter or WP-PostRatings? I haven't been able to reproduce it. If anyone is still affected by this, let me know.

I had loads of issues with this and the Theme My Login plugin, and also simply with the wp_list_pages function. On last-child pages, it returned a 500 Error, and I have no idea why.
Hopefully what fixed it for me will fix a lot of the issues for all of you.
Under the Options Page for Role Scoper, in the "Realm" section, in the "Access Types" area, I unchecked "viewing content"…worked like a charm!

If you can wait on this for another week, I have an update in the works which includes fixes for page filtering conflicts with a few other plugins. If you can't wait, I think those may be in the last development snapshot I put out. Sorry to not be of more immediate help, but I need to focus on version 1.1 development and my local copy is not ready to pass out.
The Realm switch you turned off totally disables all Role Scoper filtering on the front end.
